Not with the intention of winning or anything, but purely for entertainment purposes, akin to Stephen Colbert's "run" in 2008.

Who should his running mate be, what party's nomination should he seek, and how would he fare in the general election?

Jesse Ventura has already floated the idea of running for prez in 2016 with Stern as his running mate, and claimed that Stern "has expressed interest":

I love Howard Stern like I love Ron Paul and Bob Dylan-cultishly. He won't run though, because he does not want to release his financial info. He also has grown quite leftwing in recent years, and seems to be your standard Obama Democrat.

The guy has been all over the place politically over the years. For example: I remember in 2010, he said he would never vote for Democrat again... but by 2012 he said he was voting for Obama because Romney was a "demon".
