There isn't really an effective Republican Party in NYC. They just occasionally convince big names to run.

Chicago, obviously not. The Daleys control the city. LA certainly has its share of Republicans, but I have a feeling that they're mostly organized to try to secede from LA rather than to win elections. San Francisco has more Greens than Republicans (slight exaggeration, but only slight). There might be some Republicans in white areas of Detroit, but I don't think there are many of those left these days. If there are, they probably have a similar attitude to Republicans in LA: secession.

Minneapolis' Republican Party might as well be nonexistent. The Greens have won more elections than them in recent years.

Not really in the city of Detroit, but there is in Wayne County.  There's the 13th and 14th Districts (maybe 15th too - I don't remember).  I'm active in the 13th and know people in the 14th.  The Wayne County Republican Committee is pretty active - we just had our picnic - over 50 people.  We have a Young Republicans group - which is relatively new, but growing fast.  In terms of actually in Detroit, there's not really anything, but there's definitely a group in Metro-Detroit.

I would guess the San Fernando Valley would have some party organization.  Who knows, though.  I would never put too much faith in California Republicans.

Doesn't Dallas have a decent Republican base (it is Texas after all!!!).

Oklahoma City (which I classify as a big city) definitely does.
