Not from there, never been there, but I'd say Nassau/Suffolk is a greater divide.  Suffolk County is much longer east-west than Nassau County, and Nassau County is right next to Queens in New York City.  As for what the biggest divide in Suffolk County is, the "spine" of the island or (roughly) some line of longitude, I can't say.
